    Characterization of glucose oxidation by gold nanoparticles using nanoceria

This manuscript version is made available under the CC-BY-NC-ND 4.0 license http://creativecommons.org/licenses/by-nc-nd/4.0/Gold nanoparticles (AuNPs) can oxidize glucose, producing hydrogen peroxide and gluconic acid, which are the same products as those generated by glucose oxidase (GOx). In this regard, AuNPs are a nanozyme. Herein, a new colorimetric method is developed to understand the surface chemistry of gold nanoparticles for this oxidation reaction. The color of nanoceria is changed to yellow by the hydrogen peroxide generated during glucose oxidation. Using this assay, we find that adsorption of small molecules such as citrate does not deactivate AuNPs, while adsorption of polymers including serum proteins and high molecular weight polyethylene glycol inhibits glucose oxidation. In addition to glucose, AuNPs can also oxidize galactose. Therefore, this reaction is unlikely to be directly useful for glucose detection for biomedical applications. On the other hand, AuNPs might serve as a general oxidase for a broad range of substrates. The glucose oxidation reaction is slower at lower pH. Since the reaction generates an acid product, glucose oxidation becomes slower as the reaction proceeds. The effects of temperature, AuNP size, and reaction kinetics have been systematically studied.     Design of a scanning tunneling microscope for electrochemical applications

    A design for a scanning tunneling microscope that is well suited for electrochemical investigations is presented. The construction of the microscope ensures that only the tunneling tip and the sample participate in electrochemical reactions. The design also allows rapid replacement of the tip or sample, and enables facile introduction of auxiliary electrodes for use in electrochemical experiments. The microscope utilizes stepper motor driven approach mechanics in order to achieve fully remote operation and to allow reproducible coarse control of tip/sample spacings for electrochemical experiments. Highly ordered pyrolytic graphite images at atomic resolution in air and aqueous solutions can be obtained with this microscope

We recently reported that dispersing citrate-capped gold nanoparticles (AuNPs) in polymers, such as polyethylene glycol (PEG), can achieve such a goal because of contributions from depletion stabilization. Because AuNPs can weakly adsorb PEG to exert steric stabilization and the remaining citrate can impart charge stabilization, the extent of the contribution of depletion stabilization is unclear. In this work, we aim to dissect the contribution of each stabilizing factor. This is achieved by coating AuNPs with a layer of thiolated compound, which inhibits the adsorption of PEG and also allows for the control of surface charge. We found that depletion stabilization alone was insufficient to stabilize AuNPs at room temperature. However, when working together with other stabilization mechanisms, ultrahigh stability can be achieved. The size of both AuNPs and PEG was systematically varied, and the trends were compared to theoretical calculations.     Characteristics of Lightning Within Electrified Snowfall Events Using Lightning Mapping Arrays

    This study examined 34 lightning flashes within four separate thundersnow events derived from lightning mapping arrays (LMAs) in northern Alabama, central Oklahoma, and Washington, DC. The goals were to characterize the in-cloud component of each lightning flash, as well as the correspondence between the LMA observations and lightning data taken from national lightning networks like the National Lightning Detection Network (NLDN). Individual flashes were examined in detail to highlight several observations within the dataset. The study results demonstrated that the structures of these flashes were primarily normal polarity. The mean area encompassed by this set of flashes is 375 square kilometers, with a maximum flash extent of 2300 square kilometers, a minimum of 3 square kilometers, and a median of 128 square kilometers. An average of 2.29 NLDN flashes were recorded per LMA-derived lightning flash. A maximum of 11 NLDN flashes were recorded in association with a single LMA-derived flash on 10 January 2011. Additionally, seven of the 34 flashes in the study contain zero NLDN identified flashes. Eleven of the 34 flashes initiated from tall human-made objects (e.g., communication towers). In at least six lightning flashes, the NLDN detected a return stroke from the cloud back to the tower and not the initial upward leader. This study also discusses lightning's interaction with the human built environment and provides an example of lightning within heavy snowfall observed by GOES-16's Geostationary Lightning Mapper

    Comparison of Aquatic-Insect Habitat and Diversity Above and Below Road Crossings in Low-Order Streams

    The effects of road crossings on fish communities have been extensively studied; yet little attention has been given to macroinvertebrate communities. This study evaluated physical stream characteristics, water quality, and aquatic-insect richness from above and below road crossings of low-order streams in the Ouachita National Forest in Arkansas. Fifteen road crossings were sampled during October and November 2005. Erosion was significantly higher below road crossings than above. Sites downstream of road crossings had significantly lower pH and significantly higher turbidity than sites upstream of road crossings. Despite differences in water quality and habitat, there was no apparent difference in aquatic-insect richness from above and below road crossings based on the EPT index, suggesting that road crossings did not act as barriers to insect movement. The water-quality differences observed were well within acceptable limits and likely not biologically important

    Connecting Teachers, Students and Pre-Service Teachers to Improve STEM Pathways in Schools

    This paper presents an example of collaboration between teacher educators, pre-service teachers, current teachers and school students that had a primary aim to increase student interest in STEM activities through a MakerSpace STEM club, while improving pre-service teachers’ confidence in delivering the Australian Curriculum: Technologies. The benefit of close relationships between universities and schools provides the framework for collaborative learning opportunities for pre-service teachers and school students. University academics were facilitators in the process, managing the external grant application and wider community workshops as well as embedding the activities in the university curricula. The school teachers managed the internal delivery of the MakerSpace club and promotion of STEM activities in their schools. All worked collaboratively to provide two professional development workshops, supported by a grant from the Google CS4HS program. Outcomes of this research demonstrate a student-centred approach to digital technology education. This model of collaboration between teacher educators and schools is replicable and has a positive impact on preparing pre-service teachers to be classroom ready

    Impact of Dietary Resistant Starch on the Human Gut Microbiome, Metaproteome, and Metabolome

    Diet can influence the composition of the human microbiome, and yet relatively few dietary ingredients have been systematically investigated with respect to their impact on the functional potential of the microbiome. Dietary resistant starch (RS) has been shown to have health benefits, but we lack a mechanistic understanding of the metabolic processes that occur in the gut during digestion of RS. Here, we collected samples during a dietary crossover study with diets containing large or small amounts of RS. We determined the impact of RS on the gut microbiome and metabolic pathways in the gut, using a combination of “omics” approaches, including 16S rRNA gene sequencing, metaproteomics, and metabolomics. This multiomics approach captured changes in the abundance of specific bacterial species, proteins, and metabolites after a diet high in resistant starch (HRS), providing key insights into the influence of dietary interventions on the gut microbiome. The combined data showed that a high-RS diet caused an increase in the ratio of Firmicutes to Bacteroidetes, including increases in relative abundances of some specific members of the Firmicutes and concurrent increases in enzymatic pathways and metabolites involved in lipid metabolism in the gut

    European normative values for physical fitness in children and adolescents aged 9–17 years: results from 2 779 165 Eurofit performances representing 30 countries

    OBJECTIVE: To develop sex-specific and age-specific normative values for the nine Eurofit tests in European children and adolescents aged 9-17 years. METHODS: A systematic review was undertaken to identify papers that explicitly reported descriptive results for at least one of nine Eurofit tests (measuring balance, muscular strength, muscular endurance, muscular power, flexibility, speed, speed-agility and cardiorespiratory fitness (CRF)) on children and adolescents. Data were included on apparently healthy (free from known disease/injury) children and adolescents aged 9-17 years. Following harmonisation for methodological variation where appropriate, pseudodata were generated using Monte Carlo simulation, with population-weighted sex-specific and age-specific normative centiles generated using the Lambda Mu Sigma (LMS) method. Sex-specific and age-specific differences were expressed as standardised differences in means, with the percentage of children and adolescents with healthy CRF estimated at the sex-age level. RESULTS: Norms were displayed as tabulated centiles and as smoothed centile curves for the nine Eurofit tests. The final dataset included 2 779 165 results on children and adolescents from 30 European countries, extracted from 98 studies. On average, 78% of boys (95% CI 72% to 85%) and 83% of girls (95% CI 71% to 96%) met the standards for healthy CRF, with the percentage meeting the standards decreasing with age. Boys performed substantially (standardised differences \u3e0.2) better than girls on muscular strength, muscular power, muscular endurance, speed-agility and CRF tests, but worse on the flexibility test. Physical fitness generally improved at a faster rate in boys than in girls, especially during the teenage years. CONCLUSION: This study provides the largest and most geographically representative sex-specific and age-specific European normative values for children and adolescents, which have utility for health and fitness screening, profiling, monitoring and surveillance
